In reply, Railways were 18 for no loss at close with openers Shreyas Khonalkar (17) and Sanjay Bangar (1) at crease.
After the hosts just managed to save the first day on Sunday, making 244 for six, determined efforts by both Sriram and Karthik, who added 212 runs for the seventh wicket in 66.1 overs, brightened Tamil Nadu's chances to progress to final.
Sriram was run out pathetically when his partner R Ramkumar sent him back on second thoughts after the former had driven the ball to mid-on region. But, before Sriram could regain his crease, J P Yadav threw the ball to wicket keeper Sushil Wankhade to do the rest.
Sriram, who had occupied the crease for 554 minutes after coming at the fall of opener S Ramesh's wicket on Sunday, made 172 with the help of 17 fours and a six.
Karthik's 122 (12x4 and 4x6) was the brightest patch in the hosts innings. He made his exit when Kulamani Parida made no mistake with a catch at mid-wicket of makeshift off spinner, Yere Goud. Ram Kumar remained unbeaten on 28 (4x4, 1x6).
Brief scores: Tamil Nadu (First Innings): 481 all out in 169.2 overs (S Suresh 44, S Sriram 172, S Sharath 26, Vasudeva Das 25, Dinesh Karthik 122 and Parida 2/136, Harvinder 2/82, J P Yadav 2/66) and Railways 18 without loss.
