Chasing a modest target of 128, Victoria reached the target with 25 balls remaining.
David Hussey showed top scored Victoria innings 31 off 15 balls, smashing three successive sixes to win the game. Brad Hodge (25), Rob Quiney (29) and Aiden Blizzard (21) laid the foundations for Hussey's late assault.
Earlier, Royal Challengers got off to a flying start as Robin Uthappa (13) and Manish Pandey (39) blasted 39 runs in 28 balls.
But, McDonald's brilliant bowling spell changed the complexion of the match with four quick strikes and it needed another propping-up act from Rahul Dravid (33 off 35 balls) for the home team to reach a competitive total.
Victoria carried two points in the Super Eight stage as they had defeated Delhi Daredevils, the second team to qualify from Group D, in the first stage of the league.
Victorians are now in a strong position to reach the final.
Royal Challengers did not carry any point from the league stage and needed to win both their league matches to have a real chance to reach the last four.
Andrew McDonald was adjudged as the man of the match.
